Bovine parthenogenotes produced by inhibition of first or second polar bodies emission Biocell
Bevacqua,Romina J.; Fernandez-Martin,Rafael; Salamone,Daniel F..
Parthenogenetic embryos are an ethically acceptable alternative for the derivation of human embryonic stem cells. In this work, we propose a new strategy to produce bovine parthenogenetic embryos inhibiting the emission of the first polar body during in vitro maturation, and allowing the extrusion of the second polar body during oocyte activation. Cytochalasin B, an inhibitor of actin microfilaments, was employed during in vitro maturation to inhibit first polar body emission or during parthenogenetic activation to block second polar body emission. Only one polar body was inhibited in each strategy in order to keep the diploid chromosome set. In experiment 1, the effect of cytochalasin B on in vitro maturation of bovine oocytes was evaluated. Most oocytes...

Computer simulations for biological aging and sexual reproduction Anais da ABC (AABC)
STAUFFER,DIETRICH; OLIVEIRA,PAULO M.C. DE; OLIVEIRA,SUZANA MOSS DE; PENNA,THADEU J.P.; MARTINS,JORGE S. SÁ.
The sexual version of the Penna model of biological aging, simulated since 1996, is compared here with alternative forms of reproduction as well as with models not involving aging. In particular we want to check how sexual forms of life could have evolved and won over earlier asexual forms hundreds of million years ago. This computer model is based on the mutation-accumulation theory of aging, using bits-strings to represent the genome. Its population dynamics is studied by Monte Carlo methods.

Effects of the conditioned medium of mesenchymal stem cells on mouse oocyte activation and development BJMBR
Feng,D.Q.; Zhou,Y.; Ling,B.; Gao,T.; Shi,Y.Y.; Wei,H.M.; Tian,Z.G..
Mesenchymal stem cells (MSCs) have been reported to secrete a variety of cytokines and growth factors acting as trophic suppliers, but little is known regarding the effects of conditioned medium (CM) of MSCs isolated from femurs and tibias of mouse on the artificial activation of mouse oocytes and on the developmental competence of the parthenotes. In the current study, we investigated the effect of CM on the events of mouse oocyte activation, namely oscillations of cytosolic calcium concentration ([Ca²+]i), meiosis resumption, pronucleus formation, and parthenogenetic development. The surface markers of MSCs were identified with a fluorescence-activated cell sorter. The dynamic changes of the spindle and formation of pronuclei were examined by...

Experimental processes of hybridization in the Laminariales of French coasts. ArchiMer
Cosson, J; Gayral, P.
The research has shown that interspecific and intergeneric hybridization is possible in the Laminariales with variable rates according to the species used; these rates were determined precisely only following preliminary study where the rate of parthenogenetic development was evaluated for each particular species. Besides, the results obtained concerning the ecophysiology of the Laminariales and especially the effect of temperature and light on the fertility of the gametophytes, prompt to undertake experimental cultures of the hybrids in order to obtain algae with qualities superior to those of the parents, particularly regarding alginic acid content and growth. The experiments carried out in 1983 concern together the rearing ashore of isolated...

Genetic structure of bisexual and parthenogenetic populations of Artemia from Italian brackish-hypersaline waters ArchiMer
Nascetti, G; Bondanelli, P; Aldinucci, A; Cimmaruta, R.
Eight populations of Artemia displaying different reproduction modalities (amphigonic vs. parthenogenetic) and ploidies (diploid vs. polyploid) have been collected from Italian salt-works or brackish coastal lagoons, and their genetic structure has been investigated using allozyme electrophoresis. The results obtained on genetic differentiation showed that the Tyrrhenian bisexual samples (A. salina) are genetically homogeneous even between populations located in relatively faraway sites (Sardinia-Latium), having a moderate gene flow assisted by migratory waterfowl. The parthenogenetic samples (A. parthenogenetica) are genetically well differentiated both from the bisexual ones and among them, even when characterised by the same ploidy. Each sample presents...

Growth and developmental time in the parthenogenetic scorpion Tityus stigmurus (Thorell, 1876) (Scorpiones: Buthidae) Biological Sciences
Souza, Adriano Medeiros de; Santana Neto, Pedro de Lima; Lira, André Felipe de Araujo; Albuquerque, Cleide Maria Ribeiro de.
This study describes variation in ontogenetic development of the parthenogenetic scorpion Tityus stigmurus based on variations in size, intermolt period, and number of instars. Individuals were created under laboratory conditions (28 ± 3ºC) and subjected to similar diet regime. Comparative measurements of carapace, segment V of the metasoma, and the movable finger are used to estimate variations in size among instars. The application of Dyar rule shows a growth factor from 1.14 to 1.38 depending on the parameter analysed with intense variations in size between and within the different instars. The duration of the period from birth to adulthood was 871 days, after six moults. These data suggest that T. stigmurus has the longest post-embryonic development of...

Host Deprivation Effect on Reproduction and Survival of Wolbachia-Infected and Uninfected Trichogramma kaykai Pinto & Stouthamer (Hymenoptera: Trichogrammatidae) Neotropical Entomology
HOHMANN,CELSO L.; LUCK,ROBERT F.; STOUTHAMER,RICHARD.
The effect of temporary host deprivation on the reproduction and survival of Trichogramma kaykai Pinto & Stouthamer was determined by comparing a Wolbachia-infected (thelytokous) and an arrhenotokous line of T. kaykai when reared on Trichoplusia ni Hübner (Lepidoptera: Noctuidae) eggs. The progeny produced by both arrhenotokous and thelytokous females decreased with increasing periods of host deprivation, but arrhenotokous females produced significantly more progeny than thelytokous females regardless of the period of host deprivation (0 to 10 days). They also produced more daughters. Host deprivation did not affect brood size within arrhenotokous or thelytokous lines. However, fewer wasps emerged from hosts parasitized by Wolbachia-infected than...

Life cycle of Poratia salvator (Diplopoda: Polydesmida: Pyrgodesmidae) Rev. Bras. Zool.
Pinheiro,Tamaris G.; Marques,Marinêz I.; Battirola,Leandro D..
Even though millipedes display high plasticity in their reproductive strategies, they are poorly studied in the tropical region. The present study describes the biological cycle and reports the parthenogenetic reproduction of Poratia salvator Golovatch & Sierwald, 2000 under environmental conditions. Collections were carried out in Pantanal of Mato Grosso and in Várzea Grande, state of Mato Grosso, Brazil. The individuals were kept at room temperature in individual plastic pots. All 18 females used in the experiment reproduced parthenogenetically, resulting in a total of 699 young individuals. Of these, 18% survived until the adult phase. The lowest survival rate of P. salvator was recorded for the first developmental stage (46%). The mean duration...

Oocyte activation and preimplantation development of bovine embryos obtained by specific inhibition of cyclin-dependent kinases Arq. Bras. Med. Vet. Zootec.
Perecin,F.; Méo,S.C.; Leal,C.L.V.; Garcia,J.M..
The efficiency of bohemine and roscovitine in combination with ionomycin on parthenogenetic activation and initial embryonic development of bovine oocytes was studied. Two experiments were performed: in the first, different concentrations (0, 50, 75 or 100µM) and different exposure periods (2, 4 or 6 hours) to bohemine or roscovitine were tested for activation rates of in vitro matured (IVM) bovine oocytes, which were pre-exposed to ionomycin. The best treatments, 75µM bohemine and 50µM roscovitine, both for 6h, were used in the second experiment, in which IVM bovine oocytes were exposed to ionomycin, followed or not by bohemine or roscovitine treatment, and evaluated for nuclear status, activation rate and blastocyst development were assessed. The...

Rare sex or out of reach equilibrium? The dynamics of F-IS in partially clonal organisms ArchiMer
Reichel, Katja; Masson, Jean-pierre; Malrieu, Florent; Arnaud-haond, Sophie; Stoeckel, Solenn.
Background: Partially clonal organisms are very common in nature, yet the influence of partial asexuality on the temporal dynamics of genetic diversity remains poorly understood. Mathematical models accounting for clonality predict deviations only for extremely rare sex and only towards mean inbreeding coefficient (F-IS) over bar < 0. Yet in partially clonal species, both F-IS < 0 and F-IS > 0 are frequently observed also in populations where there is evidence for a significant amount of sexual reproduction. Here, we studied the joint effects of partial clonality, mutation and genetic drift with a state-and-time discrete Markov chain model to describe the dynamics of F-IS over time under increasing rates of clonality. Results: Results of the...

SCORPIONS, SCORPIONISM, LIFE HISTORY STRATEGIES AND PARTHENOGENESIS J. Venom. Anim. Toxins
LOURENÇO,W.R.; CUELLAR,O..
Recently public health problems resulting from scorpion stings have shown an alarming increase in various tropical and subtropical countries such as Brazil, Mexico, Tunisia and Morocco. In some regions of Brazil, particularly in the States of Minas Gerais, São Paulo, Bahia, Goiás and the Federal District, more than 6000 scorpion stings with over 100 deaths were reported during a three-year period. In this paper, we attempt to demostrate the effects of human activities on the environment and their consequences on the distribution of dangerous species of scorpions. This topic is discussed in relation to the evolutionary ecology, life history strategies and asexual reproduction of scorpions.
